2. The discovery of curcumin dates back to around two centuries ago,
when Vogel and Pelletier reported the isolation of “yellow coloring -
matter” from the rhizomes of Curcuma Longa and named it
Curcumin. Later this substance was found to be a mixture of resin
and turmeric oil. In 1842, Vogel Jr. obtained a pure preparation of
curcumin but did not report its formulas. In the decades that
followed, several chemists reported possible structures of curcumin.
However, it was not until 1910 that Milobedzka and Lampe identified
the chemical structure of curcumin as diferuloylmrthanr, or
1,6-hepatadiens-3,5-dione-1,7-bis(4-hydroxy-3-methoxyphenyl)-(1E,6E).
Further work by the same group in 1913 resulted in the synthesis of
the compound. Subsequently, Srinivasan separated and quantified
the components of curcumin by chromatography in 1953
When Mother Nature designed curcumin, she gave it the ability to
target numerous critical mechanisms essential for the development
growth and progression of most cancers. Most pharmaceutical
drugs are designed to work only a single target (i.e. a cellular receptor,
gene, protein, etc) or physiologic pathway (i.e. inflammation, glucose
metabolism, immune modulations, etc.) However, the more we learn
about cancer, the more we recognize that in most cases each
cancer is driven by a complex array of hundreds-to-thousands
of targets and pathways. It is due to this complexity that, (some
rare exceptions) scientists have been unable to discover a
“silver bullet” cure for most cancers.
Curcumin is one of the hottest plant polyphenol extracts being
researched today. The studies have been done dating back to over

5. Inducer of detoxifying enzymes
Elevating cellular levels of enzymes involved in
the phase 2 reactions. e.g.glutathione-S-
transferase (GST), glucuronosyl-transferase,
are widely recognized as an important stratergy
against carcinogenesis. Curcumin is a potent
inducer of phase 2 enzymes.
Modulator of cell signaling targets
Normal cells do not proliferate without
mitogenic stimulatory signals. However,
self-sufficiency of these growth signals has
been an accepted mark of cancer cells.
Curcumin inhibits NF-KB (nuclear factor
kappa B), MAPK (mitogen-activated protein
kinases), AP-1, (Activator protein-1), Janus
kinases and signal transducers and
activators of transcription (JAK/STAT)
pathway, Cyclo oxygenase and
Lipo oxygeneases (COX/LOX) pathways,
matrix metalloproteinases (MMPs), tumor
necrosis factor- (TNF-  ) and induces
degradation of Receptor Tyrosine Kinases

Induction of apoptosis
Curcumin induces cytotoxicity in tumor cells
by the induction of apoptosis. Curcumin inhibits
NF-KB, downregulates the expression of Bcl-2,
and Bcl-XL and upregulates the expression of
Bax and Bak in cancer cells, inhibits Akt activity
and downregulates Notch expression levels.
