Immunohistochemistry

More than 105 antibodies being used in our laboratory (RTU and Concentrated).
Determinationof optimaldilutionforprimaryantibody,antibodytitration,validationof new
markers, validationof control blocks,andchange of control blockswere alsocarriedout.
Immunofluorescence
Direct Immunofluorescence technique.
Routinely 2 IF in average.
Renal biopsy: IgG, IgM, IgA, C3, C1q, kappa and lambda .
Skin biopsy: IgG, IgM, IgA, kappa and lambda stains were done.
Special stain
Almost all stains are carried out with in-house preparationsas per the SOP which includes:
PAS, PASD, AFB TB, GMS, Jones, MT, congo red, Thio T, Alcian blue, Alcian blue PAS, Mucicarmine
stain, Giemsa stain, Gram stain, Varhoff’s elastic stain, Van gieson stain, Reticulin stain, melanin
bleach, Prussian iron stain, luxol fast blue etc.

3. Cytology (Technical Aspects) – 3 MONTHS EXPOSURE.
TEST CONDUCTED:-
PAP smear,
All body fluids,
FNACs,
Liquid based cytology-Gynec (LBC),
In-house FNAC procedure,
Some special stain,
Cell block preparation by plasma-thromboplastic method,
IHC on cell block.
OTHER LABORATORY SKILLS& ROLE PLAYS:
Handled NABL in 2014 and related documentation work as well as participated in various
lectures related to NABL held at UNIPATH.
Assisted in data entry involving patient’s complicated reports in the computer.
Trained junior technician with the help of representing some power point presentations.
Prepared our personal laboratory staining manual with my colleague under the guidance of
pathologist.
Prepared SOPs looking at some referral books including,
“Laboratory methods in histotechnology” Edna B. Prophet, Armed force institute of
pathology-1992 (U.S).
“Theory and practice of Histological techniques” byJohn D. Bancroft, Marilyn Gamble-
2007, Nottingham U.K.
Maintained lab equipment in proper condition, its maintenance and quality management
and regarding documentations (QC/PM).
Looked after the usage of lab resources and ensured steady supply of materials needed
through maintained records.
Microwave processing for quick reports.
Ensured adequate safety of area in order to prevent any possible accidents.
Demonstrate and educate students regarding different methods used to conduct
