PRODUCT INFORMATION
Introduction Recombinant Anti-Ad fiber knob x Anti-CD40 Bispecific Antibody (Fab-scFv-Fc) requires expression of
three polypeptide chains, encoding two heavy chains (Fc-containing chains) and one light chain,
respectively. The Fab of anti-Ad fiber knob antibody variable domain is fused to the Fc-knob domain with a
hinge region and the same as the scFv of anti-CD40 to the Fc-hole domain, or vice versa. The Fab-Fc and
scFv-Fc fragments can be paired via the interaction of the Fc regions. This BsAb can retarget of adenoviral
vectors to tumour cells. It can be used in cancer gene therapy to transfer a therapeutic gene into tumour
cells aiming at selective and efficient cell killing.

protein, which is a long and thin shaft. Ad fiber knob has an important role in fiber protein structure and

Description CD40 (Cluster of differentiation 40), also referred to TNFRSF5, is a 48 kDa phosphorylated glycoprotein
encoded at the chromosome of 20q13.12 and contains 277 amino acids. This type I transmembrane protein
is originally characterized on B cells, and later found on platelets, monocytes, macrophages and dendritic
cells, as well as on non-hematopoietic cells including fibroblasts, myofibroblasts, epithelial and endothelial
cells. The CD40 receptor on antigen-presenting cells of the immune system and important for mediating a
variety of immune and inflammatory processes including memory B cell development, T cell-dependent
immunoglobulin class switching, and germinal center formation. CD40 has a ligand known as CD40L or
CD154, a type II transmembrane protein, composed of a sandwich extracellular structure and belonging to
the TNF superfamily.
