SKILLS AND EXPERTISE:
--Proficient in the use of Macintosh and PC graphics, word processing and data analysis
software including the Microsoft Office Suite, Adobe Photoshop, GraphPad Prism, flow
cytometry software (Cellquest, FACS DIVA, Modfit, Flow Jo) and internet/email
programs such as Outlook, Internet Explorer, Mozilla Firefox and Google Chrome.
-Clinical Experience: Researching eligibility and consenting patients to clinical trials
and studies. Performing lung function tests and collecting and entering clinical data into
patient database, ensuring it is accurate and up to date. Querying the database for
research information when necessary. Ensuring that patient privacy is maintained and
that collaborating researchers receive the information needed to perform their
experiments.
-Processing of patient blood, pleural fluid and urine samples. (buffy coat, serum, plasma
and freezing down samples). Thawing patient samples and running biological assays
such as ELISAs, ELISPOTs, and flow cytometry.
-20 years of flow cytometric experience, working with Becton Dickinson
FACScan, FACS Calibur, FACS Canto, LSR2, Fortessa and cell sorters FACS Vantage
and FACS ARIA 3, including multicolor flow cytometry (up to 12 colours), DNA
content, BrDU and intracellular cytokine staining, and multicolour cell sorting. For the
years between 2006 and 2013, my work was mainly multicolour cell sorting (up to 8
colours).
-14 years of in vivo mouse experience including breeding, crossing and
maintaining colonies, working with transgenic and knockout animals (models of
immune deficiency, autoimmunity, inflammatory and cancer), isolation, purification and
antibody-based sorting (magnetic beads and flow cytometry) of thymus,
spleen, lymph node, bone marrow tissue and tumour infiltrating lymphocytes (TILs),
vaccination, BrDU and antibody (intraperitoneal, subcutaneous and tail vein injections)
production of bone marrow chimeric mice, and growth of human tumours in severely
immunodeficient mice. Frozen sectioning of mouse brain and immunofluorescent
staining.
-Cellular immune response assays such as ELISPOT, ELISA, lymphocyte proliferation
3. (tritiated thymidine and non-radioactive).
-Long term (2 week) in vitro B lymphocyte differentiation assay, starting with primary
murine cells, included growing primary cells on feeder layers (cell culture) and
production of some reagents such as secreted cytokines from transfected cells. Human
and mouse T cell expansion protocols using cytokines (IL-2) and activating magnetic
beads (CD3/28).
-Immunofluorescent microscopy on cytospun lymphocytes.
-Isolation of tail or cellular DNA, PCR and DNA agarose gels.
