OPTOGENETICS - OVERVIEW
Optogenetics is a method that uses light to modulate molecular events in a targeted manner in
living cells or organisms. It relies on the use of genetically-encoded proteins that change
conformation in the presence of light to alter cell behaviour, for example, by changing the
membrane voltage potential of excitable cells.
It can also be defined as a biological technique that involves the use of light to control cells in living
tissue, typically neurons that have been genetically modified to express light-sensitive ion channels.
It is a neuromodulation method that uses a combination of techniques from optics and genetics to
control and monitor the activities of individual neurons in living tissue even within freely moving
animals and to precisely measure these manipulation effects in real-time.
The key reagents used in optogenetics are light-sensitive proteins. Neuronal control is achieved
using optogenetic actuators like channelrhodopsin, halorhodopsin, and archaerhodopsin, while an
optical recording of neuronal activities can be made with the help of optogenetic sensors for calcium
(GCaMP), vesicular release (synapto-pHluorin), neurotransmitter (GluSnFRs), or membrane voltage
(arc lightning, ASAP1), Control (or recording) of activity is restricted to genetically defined neurons
and performed in a spatiotemporal-specific manner by light.
Optogenetics provides millisecond-scale temporal precision which allows the experimenter to keep
pace with fast biological information processing (for example, in probing the causal role of specific
action potential patterns in defined neurons). Indeed, to probe the neural code, optogenetics by
definition must operate on the millisecond timescale to allow addition or deletion of precise activity
patterns within specific cells in the brains of intact animals, including mammals. By comparison, the
temporal precision of traditional genetic manipulations (employed to probe the causal role of
specific genes within cells, via "loss-of-function" or "gain of function" changes in these genes) is
rather slow, from hours or days to months. It is important to also have fast readouts in optogenetics
that can keep pace with the optical control. This can be done with electrical recordings ("optrodes")
or with reporter proteins that are biosensors, where scientists have fused fluorescent proteins to
detector proteins.

PATENT SEARCH STRATEGY
Using PatSeer, we searched individual publications and then collapsed them to one member per
Invention (Simple Families). Patent families are a group of patent applications in multiple countries
which represent the same invention.
The publications included in the report are updated as of 14th
January 2019.
Summary of Search Results:
 We started with a set of 3452 search results
 We then deduplicated them to unique inventions (Families) and got 1728 Families
 We browsed through the resulting set of record to remove any record that was irrelevant to
the search topic
 We then filtered, analyzed and came across some unwanted terms/Keywords. These records
were then removed from the resulting set.
 We flagged/rated each individual record using Search Recall™ option present in PatSeer
NOTE: All analysis in this report has been done on Simple Family (one member per family) and so the
data in the charts should be construed accordingly.

The chart below shows a number of filings and grants for optogenetics. The chart was generated on
overall records (3452 Records) and the trend analysis is for the last 15 years.
Trend analysis based on the filing of priority application indicates a gradual increase in number of
applications being filed post-2013. The maximum number of patent applications (236) taking/having
priority were filed during 2016.
How we did it?
Application and patents with their respective counts were exported as CSV and were converted to a column chart
from excel.

 The chart below represents the growth of different applications of optogenetics in the last 15 years
 Research for the cure of neurological diseases like Alzheimer’s, epilepsy, seizure or depression
using optogenetics has seen an exponential increase since 2014.
How we did it?
First records were classified under different application areas. Then these were added as layer on the application
year chart. The resulting chart was exported as an image. Note that for analysis the records of 2017 and onwards
were excluded as there is a time gap of 18 months for new filings to be published.

assumes that the more recent the age of the cited patents, the more quickly one generation of

 At the current rate it can be seen that the average time technology cycle time is 5.9 years.

First records were added using a query within the Search Query option. Then from the Dashboard for the
particular query, the chart for Technology Cycle Time was downloaded.

SUMMARY

Page 2 of 43 OPTOGENETICS - OVERVIEW Optogenetics is a method that uses light to modulate molecular events in a targeted manner in living cells or organisms. It relies on the use of genetically-encoded proteins that change conformation in the presence of light to alter cell behaviour, for example, by changing the membrane voltage potential of excitable cells. It can also be defined as a biological technique that involves the use of light to control cells in living tissue, typically neurons that have been genetically modified to express light-sensitive ion channels. It is a neuromodulation method that uses a combination of techniques from optics and genetics to control and monitor the activities of individual neurons in living tissue even within freely moving animals and to precisely measure these manipulation effects in real-time. The key reagents used in optogenetics are light-sensitive proteins. Neuronal control is achieved using optogenetic actuators like channelrhodopsin, halorhodopsin, and archaerhodopsin, while an optical recording of neuronal activities can be made with the help of optogenetic sensors for calcium (GCaMP), vesicular release (synapto-pHluorin), neurotransmitter (GluSnFRs), or membrane voltage (arc lightning, ASAP1), Control (or recording) of activity is restricted to genetically defined neurons and performed in a spatiotemporal-specific manner by light. Optogenetics provides millisecond-scale temporal precision which allows the experimenter to keep pace with fast biological information processing (for example, in probing the causal role of specific action potential patterns in defined neurons). Indeed, to probe the neural code, optogenetics by definition must operate on the millisecond timescale to allow addition or deletion of precise activity patterns within specific cells in the brains of intact animals, including mammals. By comparison, the temporal precision of traditional genetic manipulations (employed to probe the causal role of specific genes within cells, via "loss-of-function" or "gain of function" changes in these genes) is rather slow, from hours or days to months. It is important to also have fast readouts in optogenetics that can keep pace with the optical control. This can be done with electrical recordings ("optrodes") or with reporter proteins that are biosensors, where scientists have fused fluorescent proteins to detector proteins.

Patent families are a group of patent applications in multiple countries which represent the same invention. The publications included in the report are updated as of 14th January 2019. Summary of Search Results:  We started with a set of 3452 search results  We then deduplicated them to unique inventions (Families) and got 1728 Families  We browsed through the resulting set of record to remove any record that was irrelevant to the search topic  We then filtered, analyzed and came across some unwanted terms/Keywords. The chart was generated on overall records (3452 Records) and the trend analysis is for the last 15 years. Trend analysis based on the filing of priority application indicates a gradual increase in number of applications being filed post-2013. The maximum number of patent applications (236) taking/having priority were filed during 2016. How we did it? First new entrants in the last 5 years in optogenetics were identified by online review and these were filtered from the Apply Filter option present. Once this was done then Summary Report option was selected. Then under “generate report for” option Applicants (under custom fields) option were selected. Parameters required for analysis were selected from the option “Patent Fields” and the resulting chart was generated and then exported as an excel file.

LEGAL QUALITY SCORE (LQ) The legal score indicate the aggressiveness and investments of the company to protect the invention and also a measure of its perceived threat to the competition. A higher LQ portrays that the invention is well covered across different geographies and reflects the patenting strategies of innovators and peculiarities in the different patenting systems providing insightful indicator of the value of patented innovations.

The patent with a higher 360° score indicates the impact to the market, technological strength, defensiveness, aggressiveness of the company. The probability of company using the patents with good 360° scored for their products, economic progress and their core strategies is higher.
