What is Thyroid?
The thyroid is a small, butterfly-shaped gland in the front of your neck. The thyroid makes the hormone that controls the way the body uses energy. These hormones affect nearly every organ in the body and control many of the body's most essential functions. It involves breathing, heart rate, weight, digestion, and moods. Without enough thyroid hormones, the body's many functions slow down (Medline plus, 2021).
Causes and Pathophysiology of Hypothyroidism
Hypothyroidism develops when the body is not able to make enough thyroid hormone due to these conditions:
. Iodine deficiency
• Hashimoto's disease is an autoimmune disorder in which the body's immune system attacks the thyroid.
• Thyroiditis
• It can be present at birth, also called Congenital hypothyroidism
• If there is the surgical removal of part or all of the thyroid.
• In case of radiation to the thyroid
• Exposure to the external radiation
• Type 1 diabetes (Autoimmune)
• Pituitary disorder or lack of iodine (Jill et al., 2021)
. Certain drugs cause hypothyroidism such as Thionamides, Iodine, and iodine-containing drugs, Amiodarone, Lithium, Minocycline (Rizzo et al., 2017)

DETECTING PSYCHOLOGICAL INSTABILITY USING MACHINE LEARNING
This document discusses detecting psychological instability using machine learning algorithms. It proposes using various machine learning models like logistic regression, decision trees, KNN, SVM, and XGBoost to classify whether an individual exhibits signs of a mental disorder based on their behaviors and thoughts. The models will be trained on datasets containing examples of symptoms and tested using metrics like accuracy, precision, recall and F1-score. Previously most research used methods like questionnaires which have validity issues, or neural networks which can overfit. The proposed system applies a narrative review methodology to analyze literature and identify an appropriate machine learning approach to help diagnose mental illness.
Meaningful (meta)data at scale: removing barriers to precision medicine research
Randomized controlled trials (RCTs) are the gold standard for evaluating therapeutics in patient populations. The data collected during RCTs include a wealth of clinical measures, biomarkers, and tissue samples – the analysis of which can lead to the approval of new medicines that improve the lives of patients. The secondary use of these data can also fuel the discovery of novel targets and biomarkers that support precision medicine, but a lack of metadata standards creates substantial barriers to reuse.
For this talk, I will discuss the challenges that arise when aggregating diverse types of data from a large number of RCTs and present a case study in how to apply (meta)data standards for the scalable curation and integration of these data into an analysis ready form.

Pan-Cancer Epigenetic Biomarker Selection from Blood Sample Using SAS®
A key focus in current cancer research is the discovery of cancer biomarkers that allow earlier detection with high accuracy and lower costs for both patients and hospitals. Blood samples have long been used as a health status indicator, but DNA methylation signatures in blood have not been fully appreciated in cancer research. Historically, analysis of cancer has been conducted directly with the patient’s tumor or related tissues. Such analyses allow physicians to diagnose a patient’s health and cancer status; however, physicians must observe certain symptoms that prompt them to use biopsies or imaging to verify the diagnosis. This is a post-hoc approach. Our study will focus on epigenetic information for cancer detection, specifically information about DNA methylation in human peripheral blood samples in cancer discordant monozygotic twin-pairs. This information might be able to help us detect cancer much earlier, before the first symptom appears. Several other types of epigenetic data can also be used, but here we demonstrate the potential of blood DNA methylation data as a biomarker for pan-cancer using SAS® 9.3 and SAS® EM. We report that 55 methylation CpG sites measurable in blood samples can be used as biomarkers for early cancer detection and classification.

As shown by AstraZeneca in nature reviews*, one third of safety failures along the drug discovery process is linked to CNS toxicity uncovered in clinical trials. To avoid this attrition, the potential neurotoxicity of any drug going through the blood brain barrier (BBB) needs to be assessed in the very early stages of new chemical entities (NCE) research. Neurotoxicity assays can be performed on the SH-SY5Y human cell line by using High-Content Screening (HCS) technologies. The present study was performed using classical 2D and 3D culture protocols. In this poster, 2D results and preliminary 3D culture results on multiple reference compounds are depicted.

The thyroid is a small, butterfly-shaped gland in the front of your neck. The thyroid makes the hormone that controls the way the body uses energy. These hormones affect nearly every organ in the body and control many of the body's most essential functions. It involves breathing, heart rate, weight, digestion, and moods. Without enough thyroid hormones, the body's many functions slow down (Medline plus, 2021).
Causes and Pathophysiology of Hypothyroidism
Hypothyroidism develops when the body is not able to make enough thyroid hormone due to these conditions:
. Iodine deficiency
• Hashimoto's disease is an autoimmune disorder in which the body's immune system attacks the thyroid.
• Thyroiditis
• It can be present at birth, also called Congenital hypothyroidism
• If there is the surgical removal of part or all of the thyroid.
• In case of radiation to the thyroid
• Exposure to the external radiation
• Type 1 diabetes (Autoimmune)
• Pituitary disorder or lack of iodine (Jill et al., 2021)

DETECTING PSYCHOLOGICAL INSTABILITY USING MACHINE LEARNING
This document discusses detecting psychological instability using machine learning algorithms. It proposes using various machine learning models like logistic regression, decision trees, KNN, SVM, and XGBoost to classify whether an individual exhibits signs of a mental disorder based on their behaviors and thoughts. The models will be trained on datasets containing examples of symptoms and tested using metrics like accuracy, precision, recall and F1-score. Previously most research used methods like questionnaires which have validity issues, or neural networks which can overfit. The proposed system applies a narrative review methodology to analyze literature and identify an appropriate machine learning approach to help diagnose mental illness.
Meaningful (meta)data at scale: removing barriers to precision medicine research
Randomized controlled trials (RCTs) are the gold standard for evaluating therapeutics in patient populations. The data collected during RCTs include a wealth of clinical measures, biomarkers, and tissue samples – the analysis of which can lead to the approval of new medicines that improve the lives of patients. The secondary use of these data can also fuel the discovery of novel targets and biomarkers that support precision medicine, but a lack of metadata standards creates substantial barriers to reuse.
For this talk, I will discuss the challenges that arise when aggregating diverse types of data from a large number of RCTs and present a case study in how to apply (meta)data standards for the scalable curation and integration of these data into an analysis ready form.

Pan-Cancer Epigenetic Biomarker Selection from Blood Sample Using SAS®
A key focus in current cancer research is the discovery of cancer biomarkers that allow earlier detection with high accuracy and lower costs for both patients and hospitals. Blood samples have long been used as a health status indicator, but DNA methylation signatures in blood have not been fully appreciated in cancer research. Historically, analysis of cancer has been conducted directly with the patient’s tumor or related tissues. Such analyses allow physicians to diagnose a patient’s health and cancer status; however, physicians must observe certain symptoms that prompt them to use biopsies or imaging to verify the diagnosis. This is a post-hoc approach. Our study will focus on epigenetic information for cancer detection, specifically information about DNA methylation in human peripheral blood samples in cancer discordant monozygotic twin-pairs. This information might be able to help us detect cancer much earlier, before the first symptom appears. Several other types of epigenetic data can also be used, but here we demonstrate the potential of blood DNA methylation data as a biomarker for pan-cancer using SAS® 9.3 and SAS® EM. We report that 55 methylation CpG sites measurable in blood samples can be used as biomarkers for early cancer detection and classification.

As shown by AstraZeneca in nature reviews*, one third of safety failures along the drug discovery process is linked to CNS toxicity uncovered in clinical trials. To avoid this attrition, the potential neurotoxicity of any drug going through the blood brain barrier (BBB) needs to be assessed in the very early stages of new chemical entities (NCE) research. Neurotoxicity assays can be performed on the SH-SY5Y human cell line by using High-Content Screening (HCS) technologies. The present study was performed using classical 2D and 3D culture protocols. In this poster, 2D results and preliminary 3D culture results on multiple reference compounds are depicted.

Cell Therapy Expansion and Challenges in Autoimmune Disease
There is increasing confidence that cell therapies will soon play a role in the treatment of autoimmune disorders, but the extent of this impact remains to be seen. Early readouts on autologous CAR-Ts in lupus are encouraging, but manufacturing and cost limitations are likely to restrict access to highly refractory patients. Allogeneic CAR-Ts have the potential to broaden access to earlier lines of treatment due to their inherent cost benefits, however they will need to demonstrate comparable or improved efficacy to established modalities.
In addition to infrastructure and capacity constraints, CAR-Ts face a very different risk-benefit dynamic in autoimmune compared to oncology, highlighting the need for tolerable therapies with low adverse event risk. CAR-NK and Treg-based therapies are also being developed in certain autoimmune disorders and may demonstrate favorable safety profiles. Several novel non-cell therapies such as bispecific antibodies, nanobodies, and RNAi drugs, may also offer future alternative competitive solutions with variable value propositions.
Widespread adoption of cell therapies will not only require strong efficacy and safety data, but also adapted pricing and access strategies. At oncology-based price points, CAR-Ts are unlikely to achieve broad market access in autoimmune disorders, with eligible patient populations that are potentially orders of magnitude greater than the number of currently addressable cancer patients. Developers have made strides towards reducing cell therapy COGS while improving manufacturing efficiency, but payors will inevitably restrict access until more sustainable pricing is achieved.
Despite these headwinds, industry leaders and investors remain confident that cell therapies are poised to address significant unmet need in patients suffering from autoimmune disorders. However, the extent of this impact on the treatment landscape remains to be seen, as the industry rapidly approaches an inflection point.

This article is key for new coming discoveries in glioma pathology, as it allows a more specific approach where more studies can be made for an early diagnosis or even, new ways to help with the treatment.
