Amlopres (Generic Amlodipine Besylate Tablets) is
used to treat high blood pressure (hypertension) by
itself or in combination with other medicines.
Lowering high blood pressure helps prevent strokes,
heart attacks, and kidney problems.
It is also used to prevent certain types of chest pain
(Angina). It may help to increase your ability to exercise
and decrease the frequency of angina attacks.

Amlopres Tablets are manufactured by Cipla Limited, India
in the strengths of 2.5 mg, 5 mg and 10 mg.

Chemical Structure

The active ingredient contained in Amlopres tablets is
Amlodipine Besylate (Amlodipine Besilate).
Each uncoated Amlopres tablet contains Amlodipine
Besylate equivalent to 2.5 mg, 5 mg or 10 mg of
Amlodipine.

Uses of Amlopres 2.5 mg, 5 mg and 10 mg Tablets:
Amlopres (Amlodipine Besylate Tablets) is used alone or together
with other medicines to treat hypertension (high blood pressure). High
blood pressure adds to the workload of the heart and arteries. If it
continues for a long time, the heart and arteries may not function
properly.
Amlodipine is also used to relieve chronic stable angina (chest pain) or
vasospastic angina (Prinzmetal's or variant angina). This medicine is
also used to lower the risk of hospitalization for angina and heart
surgery (eg, coronary revascularization) in patients with recently
documented coronary artery disease (CAD) by angiography and
without certain heart problems (eg, heart failure or an ejection fraction
less than 40%).

Uses of Amlopres 2.5 mg, 5 mg and 10 mg Tablets:
Amlodipine is a calcium channel blocker. It works by affecting the
movement of calcium into the cells of the heart and blood vessels. This
relaxes the blood vessels and lowers blood pressure, and increases the
supply of blood and oxygen to the heart while reducing its workload.

Recommended Adult Dosage
For most patients, the starting dose of Amlopres tablets
(Generic Amlodipine Tablets) for the treatment of
Hypertension is 5 mg once daily, with a maximum dose of 10
mg once daily.
Small, fragile or elderly individuals or patients with hepatic
insufficiency may be started on 2.5 mg once daily and this
dose may be used when adding Amlopres (Amlodipine
Besilate) to other anti-hypertensive therapy.

Recommended Adult Dosage
Chronic Stable or Vasospastic Angina: The starting dose of
Amlopres (Amlodipine Besilate) is 5mg to 10 mg with a lower
dose (2.5 mg) in the elderly and in patients with hepatic
insufficiency.
The recommended dose range for patients with Coronary
Artery Disease (CAD) is 5mg to 10 mg once daily. In clinical
studies, the majority of patients required 10 mg.

Contraindications

Amlopres is contraindicated in patients with a known
hypersensitivity to Amlodipine Besylate, other dihydropyridines or
any component of the tablet.

Amlodipine is transferred into human breast milk, therefore
Amlopres is contraindicated during breast-feeding.

Amlopres is also contraindicated in patients with severe
hypotension, shock including cardiogenic shock, obstruction of
the outflow tract of the left ventricle (e.g. high grade aortic
stenosis) and haemodynamically unstable heart failure after
acute myocardial infarction

Storage Instructions
Amlopres Tablets (Generic Amlodipine Besylate
Tablets) have to be stored at controlled room
temperature i.e. from 20°C to 25°C (68°F to 77°F) with
excursions permitted between 15° to 30°C (59° to
86°F).
Keep this as well as all other medicines away from
children and pets.

Warnings
Symptomatic hypotension is possible, particularly in patients with
severe aortic stenosis. However, acute hypotension is unlikely.
Worsening angina and acute myocardial infarction can develop after
starting or increasing the dose of Amlopres tablets particularly in
patients with severe obstructive coronary artery disease.
Titrate slowly in patients with severe hepatic impairment.
No studies on the effects on the ability to drive and use machines have
been performed.
Patients who experience dizziness as an adverse reaction should avoid
driving vehicles or using machines.

Possible Side Effects
Most common Amlopres (Amlodipine Besilate) side effects
are headache and edema which occurred in a dose
related manner.
Other side effects not dose related but reported with an
incidence of more than 1.0% are headache, fatigue, nausea,
abdominal pain, and somnolence

Overdosage
Overdosage might be expected to cause excessive
peripheral vasodilation with marked hypotension and
possibly a reflex tachycardia.
In humans, experience with intentional overdosage of
Amlodipine Besylate (Amlodipine Besilate) is limited.

Treatment of Overdosage
If massive Amlopres (Amlodipine tablets) overdose should occur,
active cardiac and respiratory monitoring should be instituted.
Frequent blood pressure measurements are necessary.
Should hypotension occur, cardiovascular support including elevation
of the extremities and the judicious administration of fluids should be
started.
If hypotension remains unresponsive to these conservative measures,
administration of vasopressors (such as phenylephrine) should be
considered with attention to circulating volume and urine output.
Intravenous calcium gluconate may help to reverse the effects of
calcium entry blockade. As Amlodipine is highly protein bound,
hemodialysis is not likely to be of benefit.

Amlopres Tablets During Pregnancy
Amlodipine Besylate (Amlopres tablets) has been classified by the US
FDA as Pregnancy Category C. There are no adequate and well-
controlled studies in pregnant women. Amlodipine should be used
during pregnancy only if the potential benefit justifies the risk to the
fetus.
During animal reproduction studies, there was no evidence of adverse
developmental effects when pregnant rats and rabbits were treated
orally with Amlodipine Besilate during organogenesis at doses
approximately 10 and 20-times the maximum recommended human
dose (MRHD), respectively. However for rats, litter size was significantly
decreased (by about 50%) and the number of intrauterine deaths was
significantly increased.
