COLLAGEN
Is the major insoluble fibrous protein in the
extracellular matrix and in connective tissue.
Collagen makes up approximately 30% of
the proteins in the body.
3. There are at least 16 types of collagen, but
80 – 90 percent of the collagen in the body
consists of types I, II, and III.

Triple Helix.
The triple-helical structure of collagen has
abundance of three amino acids: glycine,
proline, and hydroxyproline

damage is far advanced, any repair attempt
appears to be ineffective leading eventually

Osteoarthritis (OA) is the most common
chronic condition of the joints. It occurs
when the cartilage or cushion between joints
breaks down leading to pain, stiffness and
swelling, pain.

The diagnosis of osteoarthritis includes a
medical history and a physical examination.
These may be followed by laboratory tests,

Various findings indicate a metabolic
response of chondrocytes to deposit
collagen type III in regions of articular
cartilage, presumably as a response to
mechanical injury or other matrix damage.

network glue expressed by articular
chondrocytes.
This appears to be part of an attempt to repair
and constrain the existing collagen network of
the extracellular matrix from further damage.
This response may be triggered by molecular
or mechanical signals, resulting from the joint
trauma.

concentrated in the chymotrypsin-
extractable collagen pool from osteoarthritic
articular cartilage to assess its potential as
a biomarker of Osteoarthritis (OA)
pathogenic mechanisms.

Existe un 1er ac unido habitualmente a una fase

El ag reacciona con él. Se lava y se hace
reaccionar con otro anticuerpo que está marcado
con una enzima y tras otro lavado se revela el
ensayo siendo proporcional el producto formado a
la cantidad de ag presente en la muestra.
27. ENSAYO COMPETITIVO
Para medir antígeno.
La muestra se hace competir con un Ag
marcado frente a un Ac.
Se lava para eliminar el componente que ha
quedado libre y se revela.
La cantidad de producto formado es
inversamente proporcional al Ag de
muestra.
28. ENSAYO INDIRECTO ( mide anticuerpos)
Se fija Ag a la fase sólida.
Se hace reaccionar con la muestra donde está
el Ac.
Tras un lavado se añade Ig antiglobulina
humana (antianticuerpo) marcada con enzima.
Se vuelve a lavar y se revela.
La cantidad de producto formado es
directamente proporcional a la cantidad de
Ac presente en la muestra.
29. SDS- PAGE
SODIO DUODECIL SULFATO
POLIACRILAMIDA GEL ELECTROFORESIS
Es el método más popular para determinar el
tamaño del polipéptido en presencia de SDS.
Se utiliza un estándar de tamaño conocido y la
movilidad está determinada por el peso
molecular y la carga eléctrica de la molécula.
30. Gracias al SDS la mayoría de las proteínas
adquieren una relación CARGA/MASA
idéntica por lo que se obtiene un
fraccionamiento que obedece a:
* Diferencia de peso
* Longitud de la cadena ( tamaño)
* Forma de la proteína.

Es una técnica analítica usada para detectar

Las proteínas procedentes son separadas
por electroforesis en gel según su tamaño,

44. CONCLUSIONES
El colágeno es una de las proteínas más abundantes del cuerpo ,
es esencial para mantener la elasticidad y fuerza de las
articulaciones. Se han implementado tratamientos a base de este
para combatir la artrosis.
La importancia no solo radica en la existencia del colágeno, también
en las pruebas que se realizaron para confirmar la sobre expresión
de este en pacientes con OA.
Es muy positivo que se siga estudiando la molécula del colágeno
para poder así llegar a prevenir enfermedades como la osteoartritis
que es una enfermedad de alta prevalencia a nivel mundial y afecta
la calidad de vida de muchas personas.
Con la evidencia que el colágeno tipo III aparece en zonas donde
hay daño y trauma articular como una manera de reparar y
restringir una lesión mayor es un avance científico que puede llegar
a ayudar mucho en posibles tratamientos contra enfermedades de
las articulaciones.
