Key Point: MenACWY-CRM (Menveo™) is a meningococcal conjugate vaccine against serogroups A, C, W-135, and Y of Neisseria meningitidis. Slide Overview: MenACWY-CRM is a quadrivalent meningococcal polysaccharide-protein conjugate vaccine against serogroups A, C, W-135, and Y. The capsular polysaccharides of each of these serogroups are conjugated to a nontoxic diphtheria toxin mutant, known as CRM197. The capsular polysaccharides are each individually obtained by fermentation of N meningitidis , purified by precipitation and column chromatography, and then processed by acid hydrolysis to achieve the desired size distribution of oligosaccharides. The CRM197 protein is produced by fermentation of Corynebacterium diphtheriae and then purified by chromatography and ultrafiltration. The appropriately sized polysaccharides are activated and subsequently attached to a bifunctional hydrocarbon spacer molecule, and then the free end of the spacer is attached to the CRM197 protein to produce the Men-CRM conjugates. The drug product consists of 1 vial containing the lyophilized MenA polysaccharide conjugate and 1 vial or prefilled syringe containing a liquid solution of the MenCWY conjugates. For injection, the lyophilized MenA is reconstituted with the liquid MenCWY component. The final formulation contains the following amounts of the polysaccharide conjugates: 10 µg of MenA-CRM and 5 µg each of the MenC-CRM, MenW-CRM, and MenY-CRM conjugates.
