2. INTRODUCTION
Parkinson`s disease (PD)
affects most of the 65 or
more year old individuals.
Genes involved in this
disease are on the eye of
many researchers.
PTEN-induced putative kinase 1 (PINK 1),
parkin, and DJ-1, are genes that have being
identified to be associated wtih PD.
In order to analyze the distribution of PINK1
gene T313M polymorphisms in the Uygur and
Han populations of China, patients with PD
and healty individuals were investigated.

4. Gen PINK1
• Nombre oficial: PTEN-induced putative kinase 1.
• Contiene instrucciones para la producción de la
proteína llamada PTEN-induced putative kinase 1.
• Se cree que esta serina/treonina proteína
mitocondrial kinasa, protege la mitocondria del mal
funcionamiento durante periodos de estrés celular.
5. Polimorfismo T313M
6. GENERAL OBJECTIVE
• To investigate the association between T313M
polymorphism at exon 4 of PINK1 gene and Parkinson´s
disease in the Uygur and Han populations of Xinjiang,
China.
7. MATERIALES Y MÉTODOS
Se realizó de acuerdo con los
criterios de diagnóstico del Banco
Británico del cerebro .
Los criterios de diagnóstico para la
aparición temprana y de aparición
tardía de PD se han definido por
resonancia magnética y los
exámenes de tomografía
computarizada.
Se excluyó a los pacientes con
PD secundaria, síndromes de
Parkinson-plus, las
enfermedades del sistema
nervioso, hipertiroidismo y otras
enfermedades genéticas

10. PCR
“Técnica de laboratorio que se utiliza para la
amplificación in vitro, directa de un gen o un fragmento
de DNA, o indirecta de un RNA”
Desnaturalización del DNA para
dar hebras sencillas.
Hibridación especifica de la
hebra sencilla con un
oligonucleótido .
Replicación de la hebra sencilla
por una DNA polimerasa a partir
del oligonucleótido anterior que
actúa como cebador.
• DNA plantilla.
• Primer`s: Fragmentos de DNA cortos
(10-25pb) que hibridan con
regionesespecificas conocidas del
DNA.
• DNTp`s: deoxinucleotidos (dATP,
dGTP, dCTP, dTTP).
• Buffer.
• MgCl2.
• DNA polimerasa.
11. RFLP
RF corresponde a Restriction
Fragments (fragmentos de restricción). Son los
fragmentos de ADN resultantes del corte
mediante enzimas de restricción.
L corresponde a Length (longitud), y se refiere
a la longitud de los fragmentos de restricción.
P corresponde
a Polymorphism (polimorfismo), palabra de
origen griego que significa literalmente
"muchas formas". La longitud de algunos de
los fragmentos de restricción difiere en buena
medida entre individuos, por lo cual son
posibles muchas formas, o longitudes, de ADN.
12. RESULTADOS
• Prueba de equilibrio Hardy-Weinberg:
Mostro frecuencias genotipicas del PINK1
iguales en los dos grupos .
• Distribución de la frecuencia alélica del
T313M
13. Figure 1. Genotypes of the PINK1 gene T313M polymorphism. Lanes 3 and 5 show
homozygous mutant sequences which cannot be digested and remain as 429 bp
fragments; type T/T. Lanes 1,2,4,6 and 7 show wild-type homozygous sequences that
were digested into three fragments of 429, 265 and 164 bp; type C/T. M, 2,000 bp
marker; PINK1, PTEN-induced putative kinase 1.
14. Figure 2. Sequencing of the fourth exon of the PINK1 gene in
Parkinson's disease shows a mutation from C to T at the 938 site. The
sequence around the mutation site is shown, with the arrow pointing
to the substitution. PINK1, PTEN-induced putative kinase 1.
15. Researchers Observations Agree or disagree
Klein et al In Italy, patients with
sporadic early-on set
disease have been
reported to have relatively
high mutation rates.
Disagree
Guo et al In patients with PD, the
distribution of T313M
polymorphisms was
significantly different
between patients from the
Uygur and Han
populations, and all were
homozygous mutants.

DISCUSSION
16. DISCUSSION
Researchers Observations Agree or disagree
Guo et al In Uygur, allele frecuencies in
the PD group were not
significantly different from the
control group. While in Han
population, allele frecuencies
were signicantly different.
Disagree
Fung HC et al The mutation frecuency is wide.
PINK1 gene mutation has not
been observed in patients with
sporadic PD in the USA.

17. PERSONALCONCLUSIONS
• We think this investigation is a great step for the medical
comunity, because it allows us to know better the influences
that has the different polymorphisms of the genes in the
parkinson´s disease.
• This investigation not only shows us the mechanism of action
of the genes in our body, but the different variables, such like
age, gender, ethnicity and genetics that have an important
influence in the parkinson’s disease. Giving us a bigger
perspective for the future investigations.
18. PERSONALCONCLUSIONS
• We think it´s impresive and very interesting how a
factor like environment can affect in a mayor scale
the polymorphisms of the genes in this disease.
• Finally, like the cause of this disease is still
unknown, we think this investigation gives a little bit
of reference for more researchers to keep looking
for answers, but we continue thinking this is just the
begining for a long investigation.
