5. INTRODUCTION L-ASPARAGINASE – CELL CYCLE Relation l-asparaginase/ asparaginesynthetase. Inhibition of cellcycle. Fibroblastic and gastriccell.
6. GENERAL OBJETIVE Demonstrate that H. pylori L-asparaginase plays an important role in cell-cycle inhibition induced by BCF on cultured cells, and show that this inhibition is more pronounced in cells with low expression levels of the enzyme asparaginesynthetase
8. MATERIALES Y METODOS CULTIVO Peptones, fluidos corporales , sistemas amortiguadores. Fuentes de carbono y sales, Agar. Cepas de H. pylori CCUG 17874, 60.190, G21, G27 y suero fetal bovino Indicadores PH, agentes reductores.
21. MATERIALES Y METODOS LISIS CELULAR Destrucción de la membrana celular. Liberar proteínas y otros componentes intracelular. Lisis usando: Glucosa DetergentesSln orgánicas
22. MATERIALES Y METODOS ELISA EIA heterogéneo. Fosfatasa alcalina y peroxidasa de rábano. Tres tipos: sándwich competitivo indirecto
28. CONCLUSIONS Helicobacter pylori affects 50% of world people, for this reason its important to know about it and identify some diagnostic method that helps in a early detection. This study is important because, it provides information on the behavior of H. pylori and that information can be used to studies to identify possible treatments.
29. CONCLUSIONS L-asparaginase is the responsible factor for cell-cycle inhibition of gastric cell lines for this reason it could be used on the diagnostic of H. pylori-relateddiseases. The L-asparaginase’s role as a potential pathogenic factor could be thoroughly analysed by studying its internalisation in cells of infected patients.
