ETS1 expression in H. pylori gastritis

2. Introduction
Helicobacter pylori
● It’s a gram-negative
● Acid resistance mechanisms (ureases).
● It’s microaerophilic and its proliferation is slow.
Gastritis
● It is an acute or chronic disease of the gastric mucosa with histological
findings of inflammation.
● There is atrophy of the gastric mucosa by Helicobacter pylori
● Parietal cell death therefore decreased gastric acid secretion.
3. Transcription factors:
Helicobacter pylori can interfere with transcription factors STAT3, NF-κB, and
β-catenin, and ETS1 to mediate inflammatory response.
cag is a group of genes that encode a type IV bacterial secretion system of
which are:
● the CagA protein that activates the NF-kB pathway that induces ETS1
● CagL protein, binds to integrins on the cell surface.
ETS1 is a proto-oncogene that plays an important role in inflammatory
disorders, also on the pathogenesis of H. pylori-associated gastritis.

4. Objectives
● Explain the expression of ETS1 in gastric epithelial cells positively
regulates the inflammatory response in Helicobacter pylori-associated
gastritis
5. MÉTODOS
PCR en tiempo real
El PCR en tiempo real permite realizar el procesos de amplificación de
fragmentos de DNA y de detección de manera simultánea.
La detección se realiza por fluorescencia, al mismo tiempo que se amplifica la
cantidad de DNA sintetizado.
La emisión de fluorescencia es directamente proporcional a la cantidad de
DNA formado.
En el artículo se usó para cuantificar el RNAm del factor de transcripción ETS1.
6. Ensayo de Luciferasa: Es utilizada para estudiar la expresión génica a nivel
transcripcional.
● Se traduce el ARNm, produciendo la
enzima reportera de luciferasa.
● Luego se lisa la célula y la luciferasa
cataliza la luciferina y ATP en Oxiluciferina
la cual genera luz. Se mide con un
luminómero.
● En este caso, usaron el promotor del gen
ETS1.
● Consiste en identificar un promotor a estudiar
● se inserta upstream del gen de Luciferasa en un plásmido.
7. Western blot
-Las células y tejido fueron puestas en gel SDS-PAGE al 10% (Buffer)
-Posteriormente se transfiere a una membrana PVDF y se observa la
migración de las proteínas al polo positivo.
-las más grandes son las que menos migrarán al polo positivo
-luego para bloquear la membrana se usó BSA al 5% y se pone en suspensión
con anticuerpos específicos
-posteriormente se incubó en anticuerpos secundarios conjugados con HRP
-las proteínas fueron visualizadas usando kit de quimioluminiscencia ECL
8. Transfección de ARNsi
-Se usó RNAsi dirigido al ETS1 y RNAsi de control
-Alrededor del 50% de monocapas confluentes fueron lavadas e incubadas en
medio libre de suero y antibióticos.
-las transfecciones se realizaron usando lipofectamine 2000

research has allowed identify especific proteins and molecules.
