HDAC8
About 5% patients have
HDAC8 mutation.
Pathogenic variation of the
HDAC8 gene results in CdLS
5 (OMIM#300882) which has
an X-linked dominant
inheritance.
HDAC8 contains 11 exons and
spans approximately 243
kilobases on human
chromosome Xq13.1.
It encodes a zinc dependent
metalloproteinase, which
regulates deacetylation of
the SMC3 protein.

SDS PAGE
La SDS PAGE es una técnica en la cual se hace una electroforesis
para separar unas proteínas presentes en una muestra, las cuales
migran por peso y carga molecular.
Su uso fue para determinar la solubilidad de la proteína HDAC8
recombinante
(PAGE = polyacrylamide gel electrophoresis)
+ detergente aniónico dodecilsulfato sódico (SDS).

Una novedosa mutación sin
sentido (c.806T> G, p.I269R)
en el gen HDAC8 se identificó
como una mutación
patogénica gracias a los
estudios realizados.
Los resultados mostraron
que la nueva mutación sin
sentido fue una mutación de
Novo.

12. RESULTADO
Comparando los rasgos tipicos de pacientes
con sindrome de Cornelia de Lange, la
paciente estudiada coincidía con la baja
estatura, un bajo peso al nacer por debajo
del percentil, microcefalia, distancia amplia
de los ojos, problemas en la alimentación,
problemas intelectuales, entre otros.
Desde el punto de vista medico, este
síndrome se caracteriza por los
siguientes fenotipos:

pathogenic mutations of the HDAC8
gene could cause CdLS 5

However, there was a report of
another female patient without arched
eyebrows and synophrys, similar to the
present patient. In addition, Kline et al.
found that the facies of CdLS patients
could
change with age.

Previous studies indicated that atypical
phenotypes of female CdLS patients
with HDAC8 mutation might be due to
SXCI. In view of this, SXCI analysis of
the present patient was also carried
out and confirmed the presence of
SXCI, which could explain the atypical
phenotype of the patient.
14. CONCLUSIONS
The study allowed us to identify that the de novo mutation of the
patient in the HDAC8 gene could lead to Cornelia de Lange
syndrome, which amplified the mutations which may be the cause
of this syndrome.
The Cornelia de Lange syndrome, although it has its own
phenotypic characteristics, can be expressed in different ways and
with varying degrees of intensity. Although the phenotype is the
most important aid to identify the pathology, there are also other
parameters that help the diagnosis
