3. VITREOUSRETINOPATHY
•Rhegmatogenous retinal detachment (RRD)
occurs when a tear in the retina leads to fluid
accumulation with a separation of the
neurosensory retina from the underlying retinal
pigmented epithelium (RPE).
•This is the most common type of retinal
detachment, which can be treated with surgery.
•The major cause of failure in surgery to correct
RRD is proliferative vitreoretinopathy (PVR),
which is characterized with formation of fibrotic-
cellular epiretinal membranes (ERMs) composed
of extracellular matrix proteins and cells,
including RPE cells, retinal glial cells, fibroblasts,
and macrophages.
4. RETINA
•The retina is the most delicate
and inner layer of the eye. It
transforms the luminous
(optical) impulse into electrical
impulse.
•The alterations in the retina
may produce several visual
problems such as scotoma,
photopsia, among others. The
retina detachment is
extremely severe due to the
fact that it causes blindness
and there is no treatment.
5. MDM2
•MDM2 is a gene and a protein
that regulates the cell cycle. It
responds to p53 and it
regulates it as well. These two
have a feedback relation.
•If the p53 is too high it
stimulated the expression of
MDM2 which inhibits the
expression of p53, and
increases its degradation.
6. MDM2 VS
RETINA
•In normal circumstances, RPE is
going to regenerate. Meaning that
p53 will be active, inducing
apoptosis and inhibiting MDM2, in
old cells. On the other hand,
MDM2 will be stimulated inducing
degradation of p53 and therefor
cell survival, in young cells.
•In PVR, cells tend to form
membranes as said previously so
this might give a small hint that
MDM2 is going to be active.
Stimulating the degradation of
p53, this way, avoiding apoptosis
in cells and encouraging the
formation of membranes.
7. OBJECTIVE
The goal of this study was to determine
whether this MDM2 T309G contributes to the
development of experimental PVR.

CUANTITATIVA
PCR: REACCIÓN EN CADENA DE
LA POLIMERASA
FUNDAMENTO: El PCR es un
método que se usa para
verificar la presencia y cantidad
de una secuencia especifica en
el ADN, por medio de su
amplificación.
¿PARA QUÉ SE USÓ?: En este
estudio se usó la PCR para ver
la presencia del gen hHPRT1
(housekeeping gen) y MDM2.
10. WESTERN BLOT
FUNDAMENTO: Separación de
moléculas (proteínas) por carga
y peso molecular. Se usa una
detección por medio de
anticuerpos (primario y
secundario)
¿PARA QUÉ SE USÓ?: En este
estudio se usó para detectar la
presencia y cantidad de p53,
MDM2 y -actina.
11. ENSAYO DE
PROLIFERACIÓN
CELULARFUNDAMENTO: Este es un
método que se usa para ver la
vida celular por medio de
adición de medios de cultivos
ricos en nutrientes y por
conteo de células.
¿PARA QUÉ SE USÓ?: En este
estudio se usó para contar
células y ver la consistencia del
experimento, es decir, para ver
como el medio y los cambios
genéticos afectaron las células
en su desarrollo y crecimiento.
12. ENSAYO DE
APOPTOSIS
FUNDAMENTO: Se usó FITC
conjugado con anexina V y PI,
una combinación de sustancias
fluorescentes y marcadores
específicos para células
muertas o en proceso de
apoptosis.
¿PARA QUÉ SE USÓ?: En este
estudio se usó para ver la
supervivencia celular teniendo
en cuenta el medio en el cual
se pusieron y su mutación.
13. PVR
EXPERIMENTAL
EN CONEJOSFUNDAMENTO: Inducción de
PVR con virectomía (ambiente)
y hematoxilina eosina en corte
histológico (hematoxilina es
morada y básica, con afinidad
por lo acido y la eosina es
rosada y acida con afinidad por
lo básico.
¿PARA QUÉ SE USÓ?: En este
estudio se usó para ver el
desarrollo de la enfermedad
con las nuevas variables en un
caso in vivo, no solo in vitro.
