Este documento describe el desarrollo de una plataforma modular de vacunas para la exhibición multimérica de antígenos utilizando un modelo de ortobunyavirus. El virus de Schmallenberg se utilizó como modelo para evaluar la funcionalidad y aplicabilidad de la nueva plataforma de vacunas. Los métodos incluyeron clonación, SDS-PAGE, ELISA y RT-qPCR. 4. ● Schmallenberg virus (SBV) as a model
for a newly evolving and fast
spreading pathogen to evaluate the
functionality and applicability of our
novel vaccine platform. SBV is an
orthobunyavirus belonging to the
family Peribunyaviridae. It was first
detected in 2011, and has
subsequently caused a large epizootic
in European livestock.

5. GENERAL OBJECTIVE
Demonstrate that the immunogenicity and
vaccine efficacy of selected antigens are
markedly improved by multivalent
presentation in the newly developed LS
MPSP.
6. MATERIALS AND METHODS
El plásmido se introduce en las bacterias mediante un
proceso llamado transformación, y las bacterias que
llevan el plásmido se seleccionan mediante
antibióticos.
Las bacterias con el plásmido correcto se utilizan para
producir más ADN plasmídico o, en algunos casos, se
inducen a expresar el gen y producir proteína.
Purificar los plásmidos obtenidos de una sola colonia
de bacterias.
Replicar el ADN genético de la bacteria, replicarlas en
otras células para prepararlas y se utilizadas
posteriormente.

Se basa en la electroforesis de
proteínas.
El principio general de la
electroforesis se refiere a la
migración de partículas o solutos
cargados en un medio líquido, bajo
el dominio de un campo eléctrico.
Se realizó para la separación de
antígenos como la parte terminal

8. MATERIALS AND METHODS
Mediante la prueba ELISA se
identifica un anticuerpo o un antígeno
utilizando un antígeno o un anticuerpo
adsorbido a una superficie o fase
solida, según el caso.
Se usó un ELISA basado en proteína N
disponible comercialmente (ID Screen
Schmallenberg virus Competition
Multi-Species, IDvet) para la
detección de anticuerpos N-
específicos del SBV (virus de
Schmallenberg).
PCR es una técnica de
amplificación enzimática de
secuencias específicas de ADN. El
método consiste en aumentar un
segmento específico de AND,
usando una polimerasa.
Para la detección de ARN de SBV se
aplicó un ensayo de RT-qPCR
basado en el segmento S con un
estándar externo para la
cuantificación del ARN viral.
