and elucidate the mechanism of wheat gluten gelation

7. Métodos
Preparación de geles
• Gluten de trigo mojado se liofilizo y se molió hasta quedar en polvo
• Las dispersiones de gluten se prepararon en agua destilada y los
valores de pH se ajustaron a 7
• Se incubaron a 25,40,50,60,70,80 y 90ºC por 30 min
• las reacciones se pararon pasando las muestras a un baño de agua
con hielo
• Las muestras resultantes se liofilizaron y se usaron para las interacciones
químicas siguientes
8. Determinación de proteínas
Basado en el gel o la textura de la proteina resolubilizada
por reactivos selectivos que rompen puentes de hidrogeno,
ionices o interacciones hidrofobicas, reflejando la
correspondiente contribución de las diferentes interacciones
químicas
La concentración de proteína en los sobrenadares
se determino usando el metodo de Kjeldahl

exposed amino acid side chains and free SH groups were
available for intermolecular interactions; consequently, when
the aggregation rate is faster than the unfolding rate, a rigid

22. Conclusions
In gel formation all the bonds interaction are important to set up
many characteristics like the topics that are studied in the
rheological measurements. So its very important to know the intra
and intermolecular interactions if we want to work or do some study
for wheat gluten gel.
“The higher change in the structure is in the secondary structure
during gelation because before 50ªC we see random coil structure
and then when the temperature is higher new b-sheets are making.”
Its necessary the heating process to make a uniform gel.
23. “During heating process the content of SH free groups decrease
and disulfide bonds increase so in the electrophoresis like SDS-PAGE
we see a decrease in the HWM-GS” so we can said that if we see a
higher decrease the gel are more uniform so it have a three
dimensional wheat gluten gel network.
The freeze-dried is necessary to make this studies because with this
process we “delete” the water in many compounds and the results
will be more reliable.
