2. Proteus mirabilis
- Gram- negative bacteria that belongs to the Enterobacteriaceae bacterial family.
- It´s the main cause besides Escherichia coli, of symptomatic infections associated to the
urinary tract, such as cystistis and pyelonephritis, sometimes leading to the formation of
urinary stones
- Most UTIs (Urinary Tract Infections) associated with this bacteria are consequent to the
catheter usage
- The virulence of this microorganism is due to several factors such as: Urease and protease
production, fimbriae-mediated bacterial attachment to the uroepithelium and hemolysin

- Polymerase Chain Reaction
- PCR is a technique used to amplify a specific DNA region
- PCR is based on using the ability of DNA polymerase to synthetize a new strand that complements the
template strand
- The objective of PCR is to produce enough copies of the target DNA, to make possible the analysis

- Enterobacterial Repetitive Intergenic Consensus Polymerase Chain Reaction
- Las secuencias ERIC se encuentran en una variedad de genomas bacterianos, incluido el P.
mirabilis
- Hace posible la evaluación de la variabilidad clonal y parentesco filogenético de estos
microorganismos
- Fue utilizado para evaluar la eficiencia, fiabilidad y la habilidad discriminatoria para realizar
estudios en microorganismos, también para determinar la variabilidad genética entre los
diferentes grupos
6. Análisis Cluster
- Realiza una agrupación de los microorganismos, de acuerdo a sus características genéticas
- En este estudio, se analizaron unas cepas de Proteus estos fueron agrupados según sus
características
- Fueron organizadas en 7 grupos homogéneos (A-G)
7. Extracción de DNA
- Separación y purificación del DNA con el fin de poder estudiarlo, analizarlo o manipularlo. En estudios,
es utilizado para se analizar y diagnosticar a pacientes con enfermedades neurodegenerativas, cáncer,
infecciones.
- DNA extraído se le midió la calidad mediante muestras de gel de agarosa al 1% y luego se le realizó
electroforesis para verificar la pureza y la integridad de la muestra

8. Identificación de P. mirabilis aislados
292 muestras de orina fueron aleatoriamente recolectadas de pacientes que padecen de infecciones
del tracto urinario (Enero-Junio 2018)
P. mirabilis aislados fueron identificados mediante pruebas bioquímicas y microbiológicas:
- Producción de catalasa y ureasa*
- Producción de gas a partir de glucosa
- Motilidad*
- Producción de ácido sulfhídrico (H2S)*
- Rojo de metilo (Methyl Red. MR)
Inoculación de los P. mirabilis confirmados en trypticase soy broth con 15% de glicerol a -20°C

14. Conclusion
- Through molecular biology, it is possible to study the genetics of microorganisms such as P.
mirabilis and establish the reasons why they cause infectious diseases such as UTIs
- Studying the genome of microorganisms, is possible to determined a correct treatment plan,
avoiding situations such as antibiotic resistance
- Molecular biology allows to analyze the ways external factors can affect the functioning and
metabolism of the microorganism that causes the disease
