Este documento describe un estudio que secuenció el genoma de la bacteria Treponema pallidum pertenue, la cual causa el yaws, para desarrollar una prueba de reacción en cadena de la polimerasa (PCR) modificada que pueda detectarla. La prueba PCR existente no podía detectar muestras de pacientes con evidencia clínica y serológica de yaws. 4. OBJECTIVE

next-generation sequencing to

pallidum subspecie pertenue was
not detected using the 2015 CDC
real-time PCR assay in samples
from patients showing clinical
and serological evidence of yaws,
and to develop a modified assay
capable of detecting these
missed samples.»
5. MÉTODOS
Recolección de la
muestra:
Se recogieron 63 muestras
de lesiones ulcerativas con
una clínica consistente en
PIAN (yaws).
Extracción del DNA:
Se precipitan las proteínas
celulares para que solo
quede el DNA como
sobrenadante y se extraiga.
Se hizo para detectar la
presencia de T. pallidum,
detectando el gen tp47 por
PCRq, que esta en todas
las subespecies de esta.
6. MÉTODOS
PCRq:
Se amplifica varias veces una secuencia
especifica de ADN durante varios ciclos
repetidos por una polimerasa. Se analizan los
productos de estos ciclos para detectar la
secuencia especifica por marcadores
fluorescentes.
Se hizo para identificar la presencia de T.
pallidum en las lesiones. Las muestras positivas
se enviaron a ser secuenciadas.
7. MÉTODOS
Secuenciación del DNA:
Se hacen varias cadenas con
nucleótidos marcados con
flurocromos a partir de un primer.
Se hace una electroforesis en un
capilar para diferenciar
fragmentos por tamaño, de modo
que mientras más pequeños, más
rápido migran. Se identifica su
longitud de onda cada que migran
a través de un laser y así se
conoce la secuencia.

PCR modificada:
Diseñaron 2 nuevos primer para comparar su eficacia al
identificar la subespecie de la espiroqueta, con la
prueba preestablecida: the 2015 Centers for Disease
Control and Prevention (CDC) real-time PCR assay
“modified PCR-1” (5ʹ-GTGCGGTGAGCCCGGCGTT-
3ʹ) and
“modified PCR-2” (5ʹ-GTGAGCCCGGCGTT-3ʹ).
Se hizo para identificar más selectivamente la
subespecie pertenue del T. pallidum y así clarificar si los
ensayos ya establecidos por el ministerio eran eficaces.

14. CONCLUSION
1.The strains circulating in the Solomon
Islands had their own characteristics in their
genome sequences.This is what made the
2015 PCRq assay fail ini the detection of T.
pallidum pertenue.
2.The sequencing of the different strains of
T. pallidum pertenue should be done in each
endemic region so the diagnosis of yaws can
be well done and false-negatives can be
avoid.
