The authors' goal was to investigate the biological role
of CIRP in apoptosis by regulating pro and anti
apoptotic genes using an etoposide-induced apoptosis
model.

8. Transfección
• La transfección consiste en la introducción
de material genético externo en células
eucariotas mediante plásmidos, vectores víricos u
otras herramientas para la transferencia.
• La transfeccion se utilizó en este estudio para
implementar el gen del CIRP y sobreexpresarlo en
células previamente preparadas en un medio
contolado.

proteínas se transfieren a un filtro que se incuba

que es sujeto de prueba.

La PCR es una técnica cuyo objetivo es la amplificación directa de
un gen o un fragmento de DNA o indirecta de un RNA.
Consta de 3 etapas
1. Desnaturalizacion: Se calienta para separar las dos hebras de
DNA.
2. Alineamiento: Hibridación de hebras sencillas con primers y
DNTps.
3. Síntesis: Etapa de amplificación donde la DNA polimerasa elonga
primers utilizando molde de hebras.
Esta técnica es utilizada en el estudio para amplificar el gen de
CIRP de placenta humana e introducirlo en las células competentes.

12. Viabilidad Celular
• La viabilidad celular se refiere a la proporción de
células que sobreviven a alguna situación particular.
• En este estudio se utilizó para ver la viabilidad o no
de las células ante el tratamiento con etopósido.
• Se usó azul de triptano como colorante que permite
diferenciar células vivas de muertas.
13. Citometría de Flujo
• Es una tecnología compleja que permite analizar una
población celular proporcionando una amplia variedad de
parámetros que van desde tamaño celular hasta medidas
de densidad.
• Esta prueba fue realizada para determinar el número de
células en apoptosis y necrosis sobre la población total
de células utilizando Annexin-V junto con PI (Ioduro de
propidio).
• La prueba fue repetida tres veces de manera
independiente para confirmar datos.
