29. CONTROL DE LA PRODUCCION DE TUMORES
We found that quercetin-induced selective cell death is caused by
mitochondrial accumulation of p53 and is sufficient to prevent teratoma
formation after transplantation of hESC- or hiPSC-derived cells. Taken
together, these results provide the “proof of concept” that small-molecule
targeting of hPSC-specific antiapoptotic pathway(s) is a viable strategy to
prevent tumor formation by selectively eliminating remaining
undifferentiated pluripotent cells for safe hPSC-based therapy.
30. CONTROL DE LA PRODUCCION DE TUMORES
Follow-up in vivo studies revealed that FL118 exhibits superior antitumor
efficacy in human tumor xenograft models in comparison with irinotecan,
topotecan, doxorubicin, 5-FU, gemcitabine, docetaxel, oxaliplatin, cytoxan
and cisplatin, and a majority of mice treated with FL118 showed tumor
regression with a weekly64 schedule. FL118 induced favorable bodyweight-loss profiles (temporary and reversible) and was able to eliminate
large tumors. Together, the molecular targeting features of FL118 plus its
superior antitumor activity warrant its further development toward clinical
trials.
